Mechanistic Summary: Sarcomeric mechanical strain activates stretch-activated ion channels, stimulating vesicular trafficking and exocytosis of CLCF1 (Cardiotrophin-like cytokine factor 1). CLCF1 traverses the microvasculature to ligate CNTFR/gp130/LIFRβ on osteoblasts, upregulating STAT3/Runx2 osteogenesis, while auto-phosphorylating Akt/mTOR in myocyte satellite cells to suppress ubiquitin-proteasome proteolysis.
