At Day 180 in unguided organoid culture, deep-layer neurogenesis (CTIP2/TBR1) is robust, but non-physiologic metabolic and ER stress stimulates precocious GFAP+ astrocytic differentiation while upper-layer cortical projection neuron specification (SATB2) is arrested by 4.2 weeks relative to natural fetal neocortex ontogeny at GW 23.6.
